Ondansetron Side Effects

Ondansetron Injection is used to prevent nausea and vomiting associated with chemotherapy, radiation therapy, and surgery. It works by blocking serotonin receptors in the brain and gastrointestinal tract, which are involved in the vomiting reflex. This antiemetic is administered intravenously for rapid action, offering relief to patients undergoing treatments that induce severe nausea. Ondansetron is often a crucial component of supportive care plans in a clinical setting, enhancing patient comfort and treatment compliance.

Apotex Corporation introduces Ondansetron Injection, a potent antiemetic designed to provide effective relief from nausea and vomiting associated with chemotherapy, radiation therapy, and postoperative recovery. Delivered in convenient 4 mg per 2 mL single-dose vials, this medication ensures accurate dosing and swift preparation, supporting healthcare professionals in delivering optimal patient care. Whether in oncology, surgery, or general medicine departments, Ondansetron Injection offers a trusted solution for managing one of the most common and distressing treatment-related side effects.

Product Details and Features:

Active Ingredient: Each 2 mL single-dose vial contains 4 mg of Ondansetron, a widely respected antagonist of serotonin 5-HT3 receptors.
Packaging: Supplied as single-dose vials for assured sterility and accurate dosing, ideal for tailored patient management.
Indications: Indicated for preventing nausea and vomiting due to emetogenic cancer treatments as well as following surgical procedures.
Administration: Designed for intravenous administration, providing rapid onset of action to address acute nausea and vomiting.
Quality Manufacturing: Produced by Apotex Corporation, ensuring adherence to high-quality control standards for safety and efficacy.

Mechanism of Action:

Ondansetron functions by selectively blocking the action of serotonin (5-HT3) at its receptor sites, both peripherally on vagal nerve terminals and centrally in the chemoreceptor trigger zone of the central nervous system. By inhibiting this pathway, the medication effectively prevents the initiation of the vomiting reflex, thus alleviating nausea and vomiting associated with various medical treatments.

Warnings:

  • Hypersensitivity Reactions: Contraindicated in patients with known hypersensitivity to ondansetron or any of its components. Monitor for signs such as rash, itching, or swelling, especially following previous similar responses to 5-HT3 receptor antagonists.
  • QT Interval Prolongation: Ondansetron may cause prolongation of cardiac QT interval. Exercise caution in patients with history or predisposition to arrhythmias, or those receiving concurrent medications known to exacerbate this effect.
  • Liver Dysfunction: Patients with hepatic impairment should be monitored closely, and dosage adjustments may be necessary.
  • Serotonin Syndrome: Use with caution in patients who are also receiving serotonergic drugs, as there is a risk for serotonin syndrome.

Side Effects:

Common side effects may include:

  • Headache
  • Drowsiness
  • Constipation

Serious side effects that require immediate medical attention include:

  • Significant changes in heart rhythm (palpitations or syncope)
  • Severe allergic reactions, such as anaphylaxis
  • Symptoms of serotonin syndrome, including agitation, hallucinations, or muscle rigidity

Ondansetron Injection from Apotex Corporation is an essential tool for healthcare providers aiming to mitigate the adverse effects of treatment-induced nausea and vomiting. With its proven mechanism of action, precise dosing form, and quick onset when administered intravenously, it supports improved patient comfort and adherence to necessary medical treatments. This trusted antiemetic stands as a testament to Apotex Corporation's commitment to high-quality pharmaceutical care.

Ondansetron Injection – Antiemetic (Anti-nausea) Medication

Ondansetron injection (brand name Zofran®) is an anti-nausea (antiemetic) drug given by intravenous (IV) or intramuscular (IM) injection in hospitals or clinics. It belongs to the class of serotonin 5-HT3 receptor antagonists, meaning it works by blocking serotonin signals that trigger nausea and vomiting. The injection form is used by prescription under medical supervision.

  • Chemotherapy/radiation-related nausea: Ondansetron injection is commonly given to cancer patients before chemotherapy or radiation therapy to prevent acute and delayed nausea and vomiting caused by these treatments. (For example, doses are often administered ~30 minutes before chemotherapy infusion.)
  • Post-operative nausea: It is also used around the time of surgery to prevent or treat nausea and vomiting from anesthesia and surgical procedures. Doctors may give ondansetron just before anesthesia or during/after surgery to reduce vomiting.
  • Other uses: Ondansetron injection may be used in other situations of severe nausea (such as after radiation or in some hospitalized patients), but its main approved uses are for chemotherapy/radiation and surgery-related nausea/vomiting.

Note: Ondansetron injection is given only by a healthcare provider; the same medication is also available in oral formulations for similar indications. It is prescription-only (not over-the-counter).

Ondansetron injection (brand name Zofran® for injection) is an anti-nausea medication given by healthcare providers to prevent or treat severe nausea and vomiting. It belongs to a class of drugs called 5-HT₃ (serotonin) receptor antagonists, which work by blocking the action of serotonin in the body (serotonin can trigger vomiting). In practice, ondansetron injection is used most often in patients undergoing cancer chemotherapy or radiation therapy (especially very emetogenic drugs like high-dose cisplatin) and in surgical patients (to prevent or treat anesthesia-related nausea). For example, patients about to start chemotherapy typically receive an IV ondansetron dose about 30 minutes before treatment (with repeat doses 4 and 8 hours later if needed) and it is given just before (or sometimes immediately after) surgery in patients at risk of postoperative nausea.

How It’s Given

Ondansetron injection is available as a sterile liquid solution. It is administered intravenously (IV) or sometimes intramuscularly (IM) in a hospital or clinic setting by a nurse or doctor. Patients do not inject it themselves at home – it is given slowly into a vein (often over 2–5 minutes or so) under medical supervision. Because ondansetron can affect heart rhythm, providers usually monitor the patient’s heartbeat during or after the infusion. Typical doses for adults are in the range of 2–4 mg per administration, but the exact dose and timing depend on the situation (e.g. chemotherapy vs. surgery) and patient condition. (The medication is also available in oral tablet or dissolving-film form for other uses, but injection ensures rapid effect for acute care).

Ondansetron Dosage in Adults

Prevention of Chemotherapy-Induced Nausea/Vomiting

  • Oral:
    • 8 mg 30 minutes before chemotherapy, then 8 mg every 8 hours for 1–2 days after
  • IV:
    • 8 mg slow IV injection over 2–5 minutes immediately before chemotherapy

Prevention of Postoperative Nausea/Vomiting

  • Oral:
    • 16 mg 1 hour before anesthesia (not common in all countries)
  • IV/IM:
    • 4 mg as a single dose at the end of surgery

Ondansetron Dosage in Children

Chemotherapy-Induced Nausea/Vomiting

  • IV:
    • Children ≥6 months: 0.15 mg/kg/dose (max 8 mg/dose), repeat every 4 hours for 3 doses
  • Oral:
    • 8–12 years: 4 mg 30 minutes before, then 4 mg every 8 hours for 1–2 days
    • ≥12 years: Adult dosing

Postoperative Nausea/Vomiting

  • Usually a single dose, but sometimes repeated once if needed

Maximum Daily Dose

  • Adults: Generally, up to 24 mg/day (8 mg every 8 hours)
  • Children: Do not exceed adult dose; specific max depends on age and weight

Special Notes

  • Liver Impairment: Max 8 mg/day
  • Forms Available: Tablets, orally disintegrating tablets, oral solution, IV, IM
  • QT Prolongation: Use with caution in patients with a history of QT prolongation

This is a general guideline. Dosing may vary. Always consult a healthcare professional for specific recommendations tailored to your situation.

Ondansetron Injection – Side Effects

Common side effects: Ondansetron (Zofran) given by injection typically causes headache and mild gastrointestinal upset. Patients often report headache (≈15–20% in trials). Gastrointestinal effects include constipation or diarrhea. Fever, chills or feeling cold have also been noted. Drowsiness or fatigue is fairly common (~7–8% of patients). Because this is given by IV/IM, injection-site reactions (pain, burning, redness or swelling) can occur. Other reported mild effects include dry mouth, abdominal cramps, paresthesias (tingling), or general malaise.

Less common side effects: Some patients experience dizziness, blurred vision or light-headedness. Gastrointestinal complaints like nausea (ironic, since Ondansetron is anti-emetic) or stomach cramps may occur. Skin effects such as itching or mild rash have been reported. Overall, these side effects are usually transient and resolve once the drug is stopped or the body adjusts.

Serious side effects (seek help): Although rare, ondansetron injection can cause severe allergic reactions. Signs include rash, hives, intense itching, or swelling of the face, tongue or throat, and breathing difficulties. Ondansetron also affects heart rhythm. It can prolong the QT interval and trigger arrhythmias. Symptoms such as chest pain, palpitations, fast or slow/irregular heartbeat, dizziness or fainting should prompt immediate evaluation. Neurologic events are rare but serious: high fever, confusion, seizures, tremors or involuntary spasms have been reported. (In extreme cases serotonin syndrome can occur – characterized by agitation, hallucinations, muscle stiffness and fever – especially if combined with other serotonergic drugs.)

Other warnings: If any side effect is severe or persistent (e.g. severe headache, stiff muscles, loss of coordination, vision changes), patients should notify their doctor promptly. Because ondansetron can interact with other medications (and underlying conditions), monitoring for these side effects is recommended in a clinical setting.
